Disulphide Bond Containing Deoxyoligonucleotide, RNA/DNA Duplex Formation

Abstract

Selectivity of RNA/DNA duplex formation with disulphide bond containing deoxyoligonucleotide compared with RNA/DNA duplex formation with unmodified deoxyoligonucleotides showed that duplexes formed with disulphide bond containing deoxyoligonucleotide have much higher selectivity than duplexes formed with unmodified deoxyoligonucleotide. The Tm data of the RNA/DNA duplexes showed that duplex formed with disulphide bond containing deoxyoligonucleotide has lower Tm than that of RNA/ unmodified deoxyoligonucleotide duplex by 10 °C. However, the Tm data of the RNA/DNA duplexes showed that duplex formed with disulphide bond containing deoxyoligonucleotides melts at 48.99°C, which is quite sufficient to be used as antisense oligonucleotide or as diagnostic probe. In fact, enhance selectivity makes the disulphide bond containing oligonucleotide more specific.
