Integrated Bioinformatics Analysis of Left-Sided and Right-Sided Colon Cancer Specific Alternative Splicing Events

Background: The difference of survival associated AS events between proximal and distal colon cancer remains elusive. We aimed to make a comparison of survival associated AS events between right-sided and left-sided colon cancer and exploit the function of these splice variants served as prognostic biomarkers.
Methods: We analyzed data on 141 left-sided colon cancer (LC) and 154 right-sided colon cancer (RC) patients for whom both genome-wide AS events profiles and clinical information were available from The Cancer Genome Atlas (TCGA) project. The prognostic predictors models and splicing correlation networks for both LC and RC were evaluated using integrative bioinformatics analysis.
Finding: A total of 630 and 1398 survival associated AS events were identified in LC and RC, respectively. Most of top survival associated AS events were favorable prognostic factors, especially in LC. The area under curve (AUC) of receiver operating characteristic (ROC) for the combined prognostic predictors model in LC and RC were over 0.9, respectively. Besides, Splicing network suggested different rules of splicing factors in LC and RC.
Interpretation: Our findings provided a novel insight into explanations of prognostic variations with
primary tumor site, suggesting AS events have different contributions in the carcinogenesis of left-sided and right-sided colon cancer.
